Resveratrol falls short in health benefits, study shows

ScienceDaily (Oct. 25, 2012) ? Resveratrol, an ingredient in red wine thought to improve insulin sensitivity, reduce risk of heart disease and increase longevity, does not appear to offer these benefits in healthy women, new research at Washington University School of Medicine in St. Louis indicates.

The study, reported online Oct. 25 in Cell Metabolism, involved 29 post-menopausal women who did not have type 2 diabetes and who were reasonably healthy. For 12 weeks, half took an over-the-counter resveratrol supplement, and the rest got a placebo, or sugar pill.

"Resveratrol supplements have become popular because studies in cell systems and rodents show that resveratrol can improve metabolic function and prevent or reverse certain health problems like diabetes, heart disease and even cancer," says senior investigator Samuel Klein, MD, director of Washington University's Center for Human Nutrition. "But our data demonstrate that resveratrol supplementation does not have metabolic benefits in relatively healthy, middle-aged women."

The results were somewhat surprising because earlier studies suggested that drinking red wine lowers the risk of health problems.

"Few studies have evaluated the effects of resveratrol in people," Klein explains. "Those studies were conducted in people with diabetes, older adults with impaired glucose tolerance or obese people who had more metabolic problems than the women we studied. So it is possible that resveratrol could have beneficial effects in people who are more metabolically abnormal than the subjects who participated in the study."

Klein, the Danforth Professor of Medicine and Nutritional Science, directs the Division of Geriatrics and Nutritional Science and the Center for Applied Research Sciences. He says many people who have heard about red wine's health benefits want to take resveratrol supplements to get the benefits of red wine without consuming large amounts of alcohol. In recent years, annual U.S. sales of resveratrol supplements have risen to $30 million.

As part of the study, Klein and his colleagues gave 15 post-menopausal women 75 milligrams of resveratrol daily, the same amount they'd get from drinking 8 liters of red wine, and compared their insulin sensitivity to 14 others who took a placebo.

The team measured the women's sensitivity to insulin and the rate of glucose uptake in their muscles, infusing insulin into their bodies and measuring their metabolic response to different doses.

"It's the most sensitive approach we have for evaluating insulin action in people," he says. "And we were unable to detect any effect of resveratrol. In addition, we took small samples of muscle and fat tissue from these women to look for possible effects of resveratrol in the body's cells, and again, we could not find any changes in the signaling pathways involved in metabolism."

But if resveratrol doesn't have a health benefit, then why are red wine drinkers less likely to develop heart disease and diabetes? Klein says there may be something else in red wine that provides the benefit.

"The purpose of our study was not to identify the active ingredient in red wine that improves health but to determine whether supplementation with resveratrol has independent, metabolic effects in relatively healthy people," he says. "We were unable to detect a metabolic benefit of resveratrol supplementation in our study population, but this does not preclude the possibility that resveratrol could have a synergistic effect when combined with other compounds in red wine."

Funding for this research comes from the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) of the National Institutes of Health (NIH), and grants from DSM Nutritional Products, the Longer Life Foundation, the Japanese Research Foundation for Clinical Pharmacology, the Manpei Suzuki Diabetes Foundation and the Kanae Foundation for the Promotion of Medical Science. Mourdock: God at work when rape leads to pregnancy - KTRE.com ...

NEW ALBANY, Ind. (AP) - Indiana Republican Senate candidate Richard Mourdock said Tuesday when a woman becomes pregnant during a rape, "that's something God intended."

Mourdock, who's been locked in one of the country's most watched Senate races, was asked during the final minutes of a debate with Democratic challenger Rep. Joe Donnelly whether abortion should be allowed in cases of rape or incest.

"I struggled with it myself for a long time, but I came to realize that life is that gift from God. And, I think, even when life begins in that horrible situation of rape, that it is something that God intended to happen," Mourdock said.

Mourdock became the second GOP Senate candidate to find himself on the defensive over comments about rape and pregnancy. Missouri Senate candidate Rep. Todd Akin said during a television interview in August that women's bodies have ways of preventing pregnancy in cases of what he called "legitimate rape." Since his comment, Akin has repeatedly apologized but has refused to leave his race despite calls to do so by leaders of his own party, from GOP presidential hopeful Mitt Romney on down.

It was not immediately clear what effect Mourdock's comments might have during the final two weeks before the Nov. 6 election. But they could prove problematic. Romney distanced himself from Mourdock on Tuesday night - a day after a television ad featuring the former Massachusetts governor supporting the GOP Senate candidate began airing in Indiana.

"Gov. Romney disagrees with Richard Mourdock's comments, and they do not reflect his views," Romney spokeswoman Andrea Saul said in an email to The Associated Press. Romney aides would not say whether the ad would be pulled and if the Republican presidential nominee would continue to support Mourdock's Senate bid.

Other Republicans did not immediately weigh in. Indiana Republican Party spokesman Pete Seat referred comment to the Mourdock campaign. A spokesman for the National Republican Senatorial Committee did not immediately return a request for comment Tuesday night.

National Democrats quickly picked up on Mourdock's statement and used it as an opportunity to paint him as an extreme candidate, calling him a tea party "zealot." DNC Chairwoman Debbie Wasserman Schultz described Mourdock's comments as "outrageous and demeaning to woman" and called on Romney to take his pro-Mourdock ad off the air.

Later Tuesday after the debate, Mourdock further explained he did not believe God intended the rape, but that God is the only one who can create life.

"Are you trying to suggest somehow that God preordained rape, no I don't think that," said Mourdock. "Anyone who would suggest that is just sick and twisted. No, that's not even close to what I said."

In response, Donnelly said after the debate in southern Indiana that he doesn't believe "my God, or any God, would intend that to happen."

Along with Romney's ad, top Republicans have been flocking to Indiana as part of an effort to break open the high-stakes race for the Senate seat, currently held by veteran GOP Sen. Richard Lugar who was defeated by Mourdock in the May primary. Republicans need to gain three seats, or four if President Barack Obama wins re-election, and seats that were predicted to remain or turn Republican have grown uncertain.

Senate Minority Leader Mitch McConnell came to Indianapolis for a fundraiser Monday, and Arizona Sen. John McCain and South Carolina Sen. Lindsey Graham campaigned for Mourdock last week. New Hampshire Sen. Kelly Ayotte is due in the state Wednesday.

Romney's coattails carry special significance in deeply conservative Indiana, where Mourdock has underperformed Romney by 12 points in most public polls. Karl Rove's Crossroads GPS also has bought another $1 million of airtime in Indiana, making his group the biggest player in Indiana's Senate race. A message left for Crossroads GPS spokesman Nate Hodson was not immediately returned.

Takeaway: Erik Eckel predicts that the iPad Mini will pose no particular disruptions to enterprise environments that already have policies and systems in place to accommodate full-size iPads, iPhones, and other mobile devices.

Enterprise technology administrators need not be worried about the new Apple iPad Mini. iPad competitors, though, that?s another story. The device will certainly prove popular. Many analysts agree the new 7.9-inch computer will blow the smaller tablet market wide open. In addition to potentially cannibalizing PC sales, Apple?s new iPad Mini immediately threatens the Amazon Kindle Fire HD and Google Nexus 7 product lines.

Systems already in place

Enterprise administrators likely won?t see major impact on their networks, however. Regardless how many millions of units Apple sells, policies already in place regarding e-mail servers, bring-your-own-device, email and Internet usage, and mobile devices should accommodate all the iPad Mini users that request joining to corporate networks.

Enterprise infrastructures should be well equipped to support iPhones, larger iPads, Android-powered smartphones, and similar devices that leverage the same e-mail server configurations, firewall port settings, and personnel procedures that new iPad Minis will, too. The devices connect using the same existing technologies and operational practices.

Little terminal services impact

It?s possible some users will request terminal services or other remote access for new iPad Minis. With a 7.9-inch display that supports a 1024?768 resolution, Apple?s smaller tablet (compared to the full-size iPad with Retina Display that measures 9.7-inches diagonally and supports a 2048?1536 resolution) is 35 percent larger than a 7-inch tablet. The larger format provides significant screen real estate that will make navigating Web pages and cloud-based apps much easier than if Apple had introduced smaller 7-inch displays.

With integrated Wi-Fi connections and optional cellular data connectivity, users will find the smaller devices more portable than full-size iPads, too, but much bulkier than a smartphone that easily fits in a jacket pocket. Thus, organizations may receive some additional remote connectivity requests but it?s unlikely those requests will prove overwhelming.

Minimal security repercussions

With proper security, Internet usage and mobile device policies, and procedures now well-honed within most enterprise environments, iPad Minis should prove to be covered by existing rules and processes implemented for smartphones, laptops and other mobile computing equipment. Little should need to change operationally to accommodate the new tablet computers, which should leverage remote wiping capacities using the same systems (OWA for users connecting using Microsoft Exchange, for example) in place for iPhones and full-size iPads. Passcode requirements, too, should still apply. IT departments just need to take time to ensure users connecting iPad Minis to the corporate network understand that the new tablets fall under the same jurisdiction and regulations as other mobile devices and are properly cataloged to ensure the organization continues to effectively track all equipment connected to its systems and resources.

Thread: Walnut doors with stained glass inserts - Family Woodworking

In early 2010, I built two pairs of walnut doors and inserted stained glass panels that I made into the doors. I described that project on-line but it seems that I did not describe it in the Family Woodworking forum. If I did, I certainly can't find it here. Margaret really likes the doors and she wants a matching pair for the passageway between our kitchen and dining room. This project in now underway and I hope to complete it before the end of November. Regular reporting upon progress seemed to make the first project proceed well, so I am going to report here on the new project too -but in less detail because the majority of the steps will be much the same as in the first project.

Two weeks ago, I finished building the six stained glass panels that will be required. They have the same design as the panels in the original set of doors but are slightly smaller because the new doors will be narrower. Here is photo of one of the panels after assembly but before soldering:

The culprits, a couple of foreign tourists, had settled down on the Spanish Steps, one of Rome?s most famous landmarks. In their hands were the offending items: sandwiches.

The officers launched into action, and after much waving of hands, the tourists wrapped up their lunch again and sheepishly slouched off.

That has been the scene in Rome?s city center since Oct. 1, when a city ordinance outlawed the consumption of food and drink on the streets in areas of ??particular historic, artistic, architectural and cultural value.??

The new rules are aimed at protecting the monuments of a city so trampled by tourists it can feel like a theme park. But they have also been met with more than a little chagrin by tourists and locals, many of whom could not resist noting that the city might have larger problems at a time when Italy is skating sideways through an economic crisis.

Time Warner Cable recently became the latest brand to fall victim to a viral customer service complaint on social media ? and this time it wasn?t just your average customer posting or tweeting. It was two generations of Star Trek captains: both Picard and Kirk.

Sir Patrick Stewart, who had recently moved to Brooklyn, became frustrated with the wait time for getting cable installed in his apartment, so he took to the Twitter-sphere to voice his new-customer discontent:


Time Warner Cable responded to the post within two minutes, a fantastic response time for customer service on social media:


Hindsight is always 20/20, but the next-day re-engagement with the actor on this very public channel instead of deflecting the conversation to a more private channel (or going straight to his apartment to make sure he was watching TV asap), made the new-customer relationship even more rocky, and encouraged Stewart?s fans and other disgruntled customers, including William Shatner, to join in for support:

The take-away?
This could happen to any company at any time, so brands should be listening and prepared to respond to customer conversations on social media. And it appears that Time Warner Cable in New York City was.

Re-engaging the classically-trained, knighted actor a day later on Twitter with a cute Star Trek-related response instead of deflecting the conversation to a more private and personalized channel. or taking care of the customer?s needs as quickly as possible, was the learning experience for the brand, and a customer service lesson for us all, as social media becomes a more prominent customer service channel.

Google's Android event: 'Nexus 4,' 'Nexus 10' and more Nexus

Google's got an Android?event scheduled for Oct. 29, about a year following the announcement of the Galaxy Nexus smartphone and Android 4.0 (better known as Ice Cream Sandwich). As with every event of this magnitude, there's a host of rumors about what might be announced. Here are the best ones:

Nexus 4
Based on photos posted to Google+ by LG and Google employees, the next Google Nexus smartphone will be made by LG and called the Nexus 4.?Given that the Nexus 7 is a 7-inch tablet, we could assume that the Nexus 4 will be a 4-inch device.

The Next Web's Matt Brian says that a "source with knowledge of Google's plans" claims that the Nexus 4 will have a quad-core 1.5 GHz ?processor, 2GB RAM, 16GB storage, an 8-megapixel rear-facing camera (and 1.3-megapixel front-facing camera). According to this individual, the device will have a?a?4.7-inch display with a?1280 x 768 pixel resolution. (If that's the case, we wonder why Google didn't round up and dub the device the Nexus 5.)

Like prior Nexus devices, the Nexus 4 is expected to be among the first to run Google's latest and greatest mobile operating system, Android 4.2.

Nexus 7
The Nexus 7 has been around since late June, after being announced during Google I/O, and Google's giving it extra attention nonetheless. According to multiple reports, the tablet will now come in two 32GB versions, ?one of which will offer HSPA+ connectivity.?

Nexus 10
Google and Samsung have been working together on a 10-inch tablet, writes the Next Web's Matt Brian. This tablet will supposedly be dubbed the Nexus 10 and offer a higher resolution display than the third-generation iPad, as previously suggested by analysts.

Android 4.2
Google is expected to show off Android 4.2 during the Oct. 29 event and, according to the Next Web's Matt?Brian, this latest version of Google's mobile operating system is expected to compete directly with Apple's iOS 6 (though that's pretty much a given). Brian's mystery source says that Android 4.2 will offer new panoramic camera features, a way to access Play Store content from a widget, and multi-user support for tablets.

The more personal efforts and skill you put, the more authentic your museum is, the richer its collection, more innovative - and it becomes a pride and personal triumph on your part, your family, and participating ?organization. ?

Another?manifestation?of your success is viewers identifying themselves with the things they see in your museum - culture, livelihood, art, etc. They feel it were their museum. This is the key parameter of the former St Paul Museum. ?Students, parents, guests, conference participants felt "at home." They held high trust and confidence in what the museum offered them to see, enjoy and learn. ?There was a variety of events, a diversity of collections. Thousands - countless in fact, came with many coming back. Testimonies of pilgrims getting their wishes come true are not uncommon; so with reported cases of healing. Spirits to me are guiding signals that sometimes take the form of humans. They carry messages that lead us to the theme of our art such as in these particular cases. The denominator is goodness ? they help us seek goodness, and goodness leads us to truth ? truth that is built by strong faith other than reason.

Can we decipher messages the same way we receive communications in daily life? I say no, not always. For the message with deep meaning are not readily evident. One has to labor in order to understand it, and capture the essence of that message.

At a corner inside the former St Paul University Museum in Quezon City, where once stood an altar many years ago when the Japanese invaders converted the campus into a concentration camp, a small group of visitors bowed in deep thoughts and prayers. For a moment these pilgrims transformed the museum into a holy place.

It was like turning back the hands of time into the Second World War. Now there is peace. There was hatred, but that too, has given way to forgiveness. Despair, and now hope, pride into humility. These contrasting scenarios provide very valuable lessons of man. For man is tempered by war and mellowed by the peace that follows it. All these took place for half a century or so.

The SPUQ museum stands as a witness of the history that shaped the school. The events are the lifeblood of the museum - its walls originally the immaculate walls once stained with blood speak of peace, its pillars the original pillars that withstood the atrocities of war and the tests of the elements and time attest to endurance and posterity.

The museum is not only a repository of history; it is the abode of history. It is like Fort Santiago or the Paco Cemetery. Or the great Pyramids of Egypt, the City of the Dead of the Aztecs, Jerusalem and Rome. These museums have one thing in common: they are part of history. They are living relics that chronicle past events,? stirring nationalism while promoting brotherhood in men. They strengthen universal values and rekindle the spirit. They bring the relationship of man with his Creator closer and harmonious.

Since its opening in late 1994, many pilgrims, old and young, parents and students, city and rural folk, have brought significance to the museum. Other than being an educational institution, it has somehow earned respect for pilgrimage.

The building is a early American architecture bearing the basic designs of Greco-Roman style ? high ceiling, prominent, bare and square pillars, solid walls with small grilled windows. The entrance is unassuming, yet there is an aura of dignity that engulfs one on opening the door. For a panoramic view meets the eye, with virtually all four corners optically converging. The scene is accentuated by the massive murals depicting some chapters of the life of St. Paul, and widened by the transparency of the glass cabinets allowing the eye to roam freely.

All these no doubt contribute to the pilgrimage atmosphere. But what is revealing is the gathered information of the place coming from no less than the sisters, many of them in their seniors and living at the nearby Vigil House then. Some of the informants have already died, but the memory of the place lives. .

The senior sisters recall the place as a prayer house. ?There was an altar which was slightly located towards the left corner of the room adjacent to the backdoor.? And they would point out the place in the museum. The backdoor leads to the basement, which was used as clinic during the Japanese occupation. The wounded and the sick were led to the prayer house and to spend time meditating, praying, or just to let time pass by. On several occasions the dead were brought for the wake.

Imagine that for a period of four years, SPUQ then a novitiate and a school for elementary and high school, was made into a garrison and concentration camp, the same way the Japanese did to UST during the same period. And also to De La Salle University in Pasay. We do not know how many died but many Filipino, American and Japanese soldiers died. There were residents, foreigners, women and children who also died.

My students would ask me whenever I tell them the story if there are ghosts on the campus ? or spirits of the dead. ?Have you seen or felt their presence?? I would counter. And the conversation lengthens, creating a world of the supernatural in the process.

Anyone would believe in spirits that may make their presence felt in one way too many, depending on who is telling the story and who are listening. I for one sensed their presence on a number of occasions. The question with believing in the supernatural though is that the mind cannot decipher reality from imagination. But it is this aspect from which we build our stories and beliefs. Take this experience as an example.

In 1994 I was painting Saul on Damascus Road into the night alone. The museum was dead silent. What a conducive time to paint! Then suddenly the arm of Saul ?moved? an inch or two downward. My brush missed the outline. I made the necessary correction but this time the arm had moved upward and now I have two errors to correct. I told myself I was too tired, and left the museum for home. That night I dreamed of Saul? holding a?red robe, which he was to use to clothe the dying Christ. Early that morning I went to the museum and continued painting the arm. I fixed Saul?s right hand and put on the red robe on it. Where did the idea of the red robe come? Was it a dream or a message I got? What made his arm move? Or was it a way of getting a message across?

I remember at one time in the early part of the painting I received visitors while I was painting the sky on makeshift scaffolding. Causally they would come and take a look at my work. Sometimes they would ask me a question or two and I would obligingly give an answer without breaking my concentration. One evening a kind sister visited the museum. She stood for sometime looking at what I was doing on the scaffolding. Anyone at the top could not see well the person below. And not know when she came and had gone. What I remember was her large hat, but that crossed my mind only days later. Who was she? Where did she come from at 9 in the evening?

At one time I was painting Paradise After Rome. This time I did it at home at our front yard. It took me till dusk. A silhouette figure kept passing at the corner of my eye. I would have dismissed it but it came twice, thrice, not saying a word and not pausing. But there is semblance of the figure I was painting with the silhouette ? a bearded man and heavily built, clothed in flowing robes. The big difference though is that the man I was painting was about to be beheaded while the silhouette was roaming free, with an air of dignity and command.

The following day I changed the man on my painting. Yes, death, I realized is resurrection. So I painted Paul, the resurrected, on the day of his execution when Rome was?being?razed by Nero?s torch.



Spirits to me are guiding signals that sometimes take the form of humans. They carry messages that lead us to the theme of our art such as in these particular cases. The denominator is goodness ? they help us seek goodness, and goodness leads us to truth ? truth that is built by strong faith other than reason.

Can we decipher messages the same way we receive communications in daily life? I say no, not always. For? the message with deep meaning are not readily evident. One has to labor in order to understand it, and capture the essence of that message.

For example on the painting, The School in Ruins, which I entitled in an accompanying verse, Grow and Bloom, Grow and Bloom, an outline of a young devil cast a shadow on the burnt building. This was discovered while I was working on the dying smoke emanating from the fresh ruins. Someone almost shouted at me, Stop, stop! and then he explained. He was seeing a devil in outstretched hand hovering over the ruins. I preserved the outline. Anyone who comes to the museum today experiences the same thing the discoverer made twelve years ago. Yes, the war, the killing, the burning, the looting are works of the devil. His imprint makes us aware not to submit ourselves to evil, but rather fight it at all cost.



The community takes pride in having a museum accredited by the National Commission for the Culture and the Arts (NCCA), and the museum curator sits in one of the Commission?s sub-committees. The SPUQ Museum is also a member of the Association of Museums in the Philippines. Because of these, the school has the opportunity to take part in various national programs in health, environment, historical events, food and nutrition, and community development, to name the major events. In return, the museum is recognized for its effort. It is one of the very few school museums given such distinction.

Our own students, faculty and the whole community recognize that here in a not far, far land is a little Smithsonian, a little Gethsemane, a little Lourdes, and a little Sistine. And the same Goodness we find there is? also found here ? here at the SPUQ Museum.

Author?s Note: Prominent pilgrims to the SPCQ Museum include high government officials, leaders in the business, university professors, journalists, personalities in the entertainment world, Filipino balikbayan and their families. The Mother Superior of SPC visited the museum on her visit to the Philippines. Officials from the United Nations, ASEAN and EU on their mission to the Philippines included in their itenerary a visit to the museum. The museum celebrated its 15th year in 2010 - its last year as conceived and made fifteen years ago (1995 to 2010).

These murals shown in this article, together with three others, have been removed from their original home for 15 years soon after I left the school as professor and curator due to old age. These murals were transferred to an open area in 2011, and are now exposed to uncontrolled environmental conditions. They can be viewed near the school canteen.?The school's modernization program included a total renovation of the original museum.?

The modernization program in the last ten years displaced the priceless botanical garden, a natural gene bank of more than 200 species of plants which were studied and?documented?by one of the country's leading biologists, Dr Anselmo S Cabigan. ?The garden had two large greenhouses modern even with today's standard. In the late nineties the garden evolved into an Ecological Sanctuary. ?Semblance of wildlife took shape with?resident?and?transient?organisms forming food chains that linked up into food webs and natural cycles ?which are indicators of a functioning ecosystem.
A series of articles in the Paulinews became a valuable reference of the then College.
In the years that followed with a boom in infrastructures, especially the school's modern buildings blocking the sun, eating out every inch of open space, the EcoSanctuary became a minuscule park of benches and?potted?plants and kiosk. ?

And the same God we find there is also found here ? here at the SPCQ Museum.

Discovery of biological energy-sensing switch could have broad implications for biology and medicine

ScienceDaily (Oct. 21, 2012) ? Biochemists at The Scripps Research Institute (TSRI) have discovered a genetic sequence that can alter its host gene's activity in response to cellular energy levels. The scientists have found this particular energy-sensing switch in bacterial genes, which could make it a target for a powerful new class of antibiotics. If similar energy-sensing switches are also identified for human genes, they may be useful for treating metabolism-related disorders such as type 2 diabetes and heart disease.

"This discovery adds a new dimension to our understanding of how cells sense and manage their energy levels, which is one of the most important processes in biology," said the study's senior author, Martha J. Fedor, a professor the departments of Chemical Physiology and Molecular Biology and a member of the Skaggs Institute for Chemical Biology at TSRI.

The findings are described online ahead of print on October 21, 2012, in the journal Nature Chemical Biology.

A Fuel Sensor

This type of gene-switching sequence is known as a riboswitch because it appears on the strand of ribonucleic acid (RNA) that is first transcribed from a gene's DNA. Unlike other known riboswitches, which have relatively limited functions, this one acts as a sensor for the basic molecular fuel that powers all living cells and controls many genes.

The newly discovered riboswitch detects a small molecule known as adenosine triphosphate (ATP), the standard unit of chemical energy in all known organisms on our planet. Scientists had thought that cells use only large and relatively complex proteins to sense these all-important energy molecules and adjust cell activities accordingly. No one had found ATP sensors among riboswitches, which can alter cell activity at a more fundamental level -- usually by interrupting a gene's transcription from DNA.

Moreover, previously described riboswitches are relatively simple feedback sensors that affect narrow metabolic pathways. Most of them merely sense and adjust the expression rate of their own host gene. "This is the first riboswitch that is known to be involved in global metabolic regulation," said Fedor.

In recent years, the Fedor team had found hints that such a riboswitch could exist. Many RNA sequences with possible riboswitch activity had never been characterized, and several riboswitches in bacteria sense molecules that are closely related to ATP. Fedor and a graduate student in her laboratory, Peter Y. Watson, therefore set out to find bacterial riboswitches that could indeed sense ATP.

Caught in the Act

The task was more challenging than it might have seemed. Watson could not simply expose suspected riboswitches to ATP and see which ones stuck best to the energy molecules. ATP is present in high concentrations in cells, and its interactions with its known protein sensors are necessarily fleeting, low-affinity affairs. Interactions with a riboswitch would be expected to look the same. "Such interactions are really too weak to be detected using traditional methods," Watson said. But he found evidence that an RNA interaction with an ATP-like molecule would occur in a way that allows the brief coupling to be caught in the act -- using a burst of ultraviolet radiation, which can create a strong chemical crosslink between two molecules.

In this way, he discovered a stretch of apparent ATP-binding RNA known as the ydaO motif. Watson performed structure-mapping analyses of ydaO to confirm that it binds to ATP and to determine precisely where it binds. Attaching ydaO to a "reporter" gene, he found that in bacterial cells, the reporter gene's expression level stayed low when ATP levels were normal and rose sharply when ATP levels dropped -- as would be expected if ydaO is really an ATP-sensing riboswitch. Even in unaltered cells of a test bacterium, B. subtilis, levels of the genes that normally contain the ydaO motif rose and fell in the same way in response to changing ATP levels.

The ydaO motif occurs in the large subset of bacteria known as gram-positive bacteria. Across these bacterial species, it has been found, to date, on 580 separate genes. "These ydaO-regulated genes encode proteins that have a wide variety of functions, from cell wall metabolism to amino acid transport," Watson said. "It makes sense that a riboswitch in control of such disparate processes would be responding to a central metabolite such as ATP."

New Possibilities

The finding has basic scientific importance because it is the first known example of a riboswitch that binds ATP; it is also the first known riboswitch that has such broad regulatory functions. "It opens up the possibility that RNA switches are involved in the general regulation of metabolism," said Fedor.

The fact that ydaO motifs serve as "off-switches" for key bacterial genes also makes them a potential target for new antibiotics. "Hitting these riboswitches with a small-molecule, ATP-mimicking drug so that they can't turn on genes that promote bacterial growth and survival could be a viable approach," said Fedor.

Her laboratory will now search for other ATP-sensing riboswitches in bacteria and in higher organisms, including humans. A human ATP-sensing riboswitch, if targeted appropriately by drugs, might be able to alter cell activity in ways that help treat common metabolic disorders. Type 2 diabetes, which presently affects several hundred million people worldwide, is known to feature the improper regulation of ATP levels in cells.

Funding for the study, "The ydaO motif is an ATP-sensing riboswitch in Bacillus subtilis," was provided by the Skaggs Institute for Chemical Biology at TSRI.

That Superman legal decision means we'll see Justice League - Blastr

Now that Warner Bros. Pictures is legally able to keep using the character of Superman, plans for the long-awaited Justice League movie are about to move faster than a speeding bullet.

As a result of yesterday's major legal victory for the studio, in which a judge ruled that the heirs of Superman co-creator Joseph Shuster were not able to win back the rights to the character, a source has told the Los Angeles Times that Warner Bros. plans to accelerate the timetable for getting the DC superhero team onto the screen.

If the studio had lost the case, it would not have been able to produce any movies, comics or TV show featuring key elements of the Superman mythology after 2013 unless it negotiated a new deal with the estates of Shuster and Jerry Siegel, Superman's other "father."

With that in mind, the studio has been hesitant about ramping up the development of the Justice League movie, in which Supes would of course be a major character. But now that the path is clear, the studio intends to begin casting and finding a director (a script has already been written). If all goes well, production should begin next year and the movie will be in theaters in the summer of 2015?where it will go head to head with The Avengers 2 for a real clash of the titans.

But now the work begins: who will get this thing on the screen and which actors/heroes will appear in it? With Christopher Nolan and Ben Affleck both passing on being involved, Warner Bros. has to find someone who can deliver the epic goods.

As for the actors, it's presumed that Henry Cavill will play Superman after making his debut next year in Man of Steel. As for Batman, Wonder Woman, the Flash, Green Lantern and whoever else makes up the team...it's all yet to be determined.

One thing's for sure though: if Justice League is a hit, the characters will spin off into their own movies. It's the exact opposite of the Marvel strategy of individual movies leading to one big team-up, but if works, we'll be seeing the DC Universe on the screen for years to come.

Who should direct Justice League and who should be in it? The speculation starts now!

Fujitsu has just revealed its Windows 8 lineup for the Japanese market, and top billing goes to the new "Floral Kiss" Ultrabook, which the manufacturer claims was built "under the direction" of its female employees in order to entice an equally female audience. At heart, it's just a regular Core i5 notebook with a 500GB hard drive, but the womanliness is all in the presentation. There are subtle color schemes like "feminine pink" and "luxury brown" to choose from and every laptop comes with pre-installed Windows 8 apps including a digital scrapbook for collecting website bookmarks, a diary and a daily horoscope checker. This almost oppressively enticing bundle will hit stores on November 2nd, with some sort of premium designer version arriving a few weeks later. As for the exact price, that'll be determined by retailers in Japan and by how good your husband is at haggling.

How flick knife thumbs help Japan's rare fighting frogs

ScienceDaily (Oct. 18, 2012) ? Combat-ready spikes which shoot from fingers sounds like the weaponry of a comic book hero, but a Japanese scientist has found exactly this in a rare breed of frog. The discovery, which is published in the Journal of Zoology, reveals how the Otton frog uses spikes which protrude from a false thumb for both combat and mating.

The study, conducted by Dr Noriko Iwai from the University of Tokyo, focused on the Otton frog (Babina subaspera), whose habitat is the Amami islands of Southern Japan. Unlike most other frogs the Otton has an extra digit-like structure, a trait it shares with the five-fingered Hypsiboas rosenbergi frogs of Latin America.

"Why these 'fifth fingers' exist in some species remains an evolutionary mystery, but the extra digit of the Otton is in fact a pseudo-thumb," said Dr Iwai. "The digit encases a sharp spine which can project out of the skin, which fieldwork demonstrates is used for combat and mating."

Dr Iwai has studied the rare frogs since 2004 in order to understand the species' distribution, breeding habits and range; all factors which will contribute to any conservation strategy. Once she began exploring how the Ottons use their pseudo-thumbs Dr Iwai discovered that while both males and females had the spike, it was only used by males.

Males were found to have larger pseudo-thumbs than the females and Dr Iwai believes that the spikes evolved for anchoring to the female, known as amplexus, the Latin for embrace, during mating.

"While the pseudo-thumb may have evolved for mating, it is clear that they're now used for combat," said Dr Iwai. "The males demonstrated a jabbing response with the thumb when they were picked up, and the many scars on the male spines provided evidence of fighting."

The conditions on the Amami islands make combat, and the need for weaponry, a key factor for the frogs' mating success. Individuals fight over places to build nests, while the chances of a male finding a mate each night are rare, thus the ability to fight off competitors may be crucial.

Perhaps unfortunately, the comic book hero image is slightly dented by the frogs' fighting style. Rather than dueling with thumb spikes the males wrestle each other in an embrace, jabbing at each other with the spines. This fighting style helps confirm the theory that the spines were original used for embracing mates.

"More research is needed to look at how the pseudo-thumb evolved and how it came to be used for fighting," concluded Dr Iwai. "The thumbs use as a weapon, and the danger of the frogs harming themselves with it, makes the Otton pseudo-thumb an intriguing contribution to the study of hand morphology."
